I'll try to limit myself to things with some mainstream appeal that's iPod-friendly (because I'll assume you don't want weird experimental stuffs)
Are you looking for recommendations of things you might buy? Or free-download only?
Alice Cooper - IMO the best 70s rock band. Their later stuff (when the band broke up and the singer took their name for his own) is more metal but their early stuff is just pure rock awesomeness. I think a good starter album would be Love It To Death.
The Avalanches - really fantastic sample-based stuff. Go look up the videos for Since I Left You and Frontier Psychologist if you don't believe me! (they only have one album so far, sadly)
Devo - they're more than just the "Whip It" band! I wholeheartedly suggest the album Q: Are We Not Men? A: We Are Devo
Electric Six - puts the "easy" back in "cheesy" (yes, I know that doesn't work) Most famous for the songs Danger High Voltage and Gay Bar.
They Might Be Giants - This band is a lot of fun, my absolute number one favorite for quite a while. Their most "beginner friendly" album is Flood. They've been long described as "nerd rock" but they've only recently accepted that label. They also have a free podcast.
YUKI - (Isoya Yuki) my favorite J-pop artist. She's fun, she's crazy, she's awesome. I don't know if you can find her stuff on iTunes or not; I only own two CDs of hers and they were kind of a pain to get, as is the case of many Japanese artists...
If you're looking for free stuff, I would heavily recommend:
Lemon Demon has a decent number of downloads available. Funny stuff! Awesome stuff!
Logan Whitehurst & the Junior Science Club - especially Earth is Big. (If you like that, you should buy Goodbye, My 4-Track)
Ytcracker's NerdRap Entertainment System - some of the best Nerdcore hip-hop out there.
I assume that the best songs for the iPod are the songs you want to hear while you're out and about more than sitting alone LISTENING. But if this is your main source of music, I'll recommend some other stuff I like.
POTENTIALLY TOO WEIRD FOR FUN IPOD TIMES BUT SHOULD BE LISTENED TO ANYWAY:
Godspeed You! Black Emperor - (formally with the exclamation point after "Emperor") really awesome "post-rock" songs are long, and build epically
Hedwig and the Angry Inch - the soundtrack to my favorite musical! I prefer the original cast recording but the movie version is fine too? It's rock! With meaning! This would be listed above but the album is SO GOOD that when I play it, I end up stopping whatever I'm doing so I can just concentrate on the music. AMAZING FANTASTIC SUPER-GREAT!
Nine Inch Nails - often too introspective to make for "music on the go." The Downward Spiral is essential music of depression? Pretty Hate Machine is really good too. The Fragile is probably the most beautiful NIN release but is definitely not the kind of music you play in the car.
Sigur Ros - Icelandic "post-rock" generally fairly soft, yet deep music. I don't know how to explain it better than that.
I own... a lot of music. Of many varieties. I'm not too great at cutting it away to "the best" but hopefully this helps!

It all depends on what you're into.
Fun music: The B-52's are great goofy rock (Love Shack, Rock Lobster). I'd suggest starting with Cosmic Thing.
Relaxing music: AIR is a French electronic band with some 70s influences. it's pretty laid back, but they have more high energy stuff, too. Start with their newest album, Love 2 (There's no Love 1, so don't worry). For relaxing yet quirky old songs, Ultra Lounge has collections of some great vocal and (mainly) instrumental songs from the 50s/60s thereabouts. Mondo Exotica is my favourite (and it has a GREAT name). Lounge music might be too weird for some, but it's great fun.
Pop/Rock: Sheryl Crow' self titled album is pretty ecclectic and worth a buy.
Jagged Little Pill by Alanis Morissette can be fairly "angry" at times (for example, the song, "You Oughtta Know"), but other songs ("Ironic," "Head Over Feet," "Hand in my Pocket") are nicer.

Check these people out for something a little different to what you'd normally listen to -
Anything by 'Dogg Master' ..especially track called Pop Lock Funk.
Check for a guy called Tryezz on youtube too, he does a very sort of 'chillout/funk' kinda sound.
Not sure if that's what you'd listen to but it's certainly something that doesn't get suggested much round here ..other than by me, lol.
